MOTS-c (Mitochondrial Open Reading Frame of the Twelve S rRNA type-c) is a 16-amino-acid peptide encoded directly within mitochondrial DNA. First described in 2015 by Dr. Changhan David Lee and colleagues at the University of Southern California, it belongs to a unique class of mitochondrial-derived signaling peptides. Unlike conventional peptides encoded by nuclear DNA, MOTS-c functions as a communication molecule that conveys signals from mitochondria back to the nucleus.
Its biological activity is primarily linked to activation of the AMPK (AMP-activated protein kinase) pathway and modulation of the folate-methionine metabolic network, both of which play central roles in cellular energy regulation. Research published in *Cell Metabolism* identified MOTS-c as a potential “exercise mimetic” because it stimulates several metabolic adaptations normally associated with physical activity, including enhanced glucose utilization, increased fatty acid oxidation, and improved mitochondrial biogenesis.
